アトピー病因遺伝子群の構造生物学的解明と環境中でのオーダーメイド治療予防への応用

研究概要	(1)解明されたアトピーの病因遺伝子群の構造生物学的解明と機能異常との関連を解明した。明らかになった多病因遺伝子群のコードするタンパクとその機能を分析した。すなわち、アレルギーの発症機構を(1)抗原プロセッシングおよびHLAクラスII抗原による抗原ペプチド提示、(2)IL-12の産生低下、(3)Th2ヘルパーT細胞の偏った活性化とIL-4、IL-5などのサイトカインの産生亢進、(4)B細胞からのIgE産生亢進、(5)好酸球、好塩基球の活性化と種々の炎症性メディエーターの産生亢進の一連の過程と、このうち明らかになった病因遺伝子がコードするタンパクを分子遺伝学的並びに構造生物学的に解析した。このうち(1)抗原特異的T細胞クローンの樹立と構造生物学:Derf1あるいはβラクトグロブリンに過敏性のある患者からクローンを樹立し、DR抗原を中心にHLA抗原と抗原ペプチドとの結合を核磁気共鳴NMRなどを用いて構造生物学的に解析した。またmutation experimentにより、核となるアミノ酸と塩基配列を決定した。(2)抗原提示細胞からのIL-12遺伝子の発現:IgE産生亢進の理由の1つをIL-12遺伝子発現低下に求め得る可能性があることから(申請者らの成績)、IL-12遺伝子の調節領域およびcoding領域の塩基配列を決定し、その異常を分子遺伝学的及びNMRを用いて構造生物学的に明らかにした。 (2)解明された遺伝子とコードされるタンパクの異常をもとにして多様な病因・病態に対応したオーダーメイド治療・予防への応用を検討した。すなわち(1)βラクトグロブリンを改変したミルクの作成(2)IL-18レセプター結合低分子の解明を用いた抑制系の修復である。
