研究実績の概要
新規に開発された中性子捕捉療法用熱中性子捕捉化合物であるp-Boronophenylalanine-10B (BPA)のαアミノアルコール体p-Boronophenylalanol(BPA-o1)が、固形腫瘍内全腫瘍細胞(P+Q)細胞とQ細胞の両方の感受性を共に高め、TPZと低温度温熱処置との併用でQ細胞の感受性をさらに高めることができ、有望な中性子捕捉化合物の一つであることが明らかになった。
また放射線感受性に大きな影響を与えるとされる腫瘍細胞のp53 statusとQ細胞の感受性との関係については、p53 statusがwild typeであろうとmutation typeであろうと、Q細胞とP+Q細胞との間の放射線感受性の差は、ほぼ変化せず一定であり、腫瘍治癒の視点から見るとQ細胞の感受性を上昇させP+Q細胞との間の感受性の差を縮小させる努力がp53 statusにかかわりなく必要であることも明らかになった。他方、中性子捕捉療法においては、固形腫瘍内では熱中性子捕捉化合物である10B化合物由来の10BはP細胞によく集積しQ細胞には集積しにくいという特性が認められるが、この特性はp53 mutation-type腫瘍よりもp53 wild-type腫瘍でより顕著に認められ、Q細胞とP+Q細胞との間の感受性の差を縮小させるという点から見ると、p53mutation-type腫瘍の方が中性子捕捉療法の良い適応となることが示された。p53 statusが変異しやすいとされる悪性腫瘍の制御には中性子捕捉療法が有用である事が示唆された。更に、生体還元物質や低温度温熱処置との併用がp53 wild-type腫瘍内のQ細胞を増感し得るという事実を我々はすでに明らかにしてきたが、p53 mutation-type腫瘍においても同等に有効であり、この併用処置による増感効果は、p53-independentであることも明らかになった。
