ヒトIgA腎症の病因と病態のゲノミックスとプロテオミックス

研究概要	1)ヒト糸球体cDNAライブラリーの6,000クローンの塩基配列を決定し、遺伝子バンクと遺伝子データベースを作成した。それらの遺伝子の中にはこれまで報告されている糸球体特異的遺伝子、nephrin、podocin、synaptopodin、podocalyxinなども含まれていた。 2)糸球体に発現している遺伝子を正常と疾患でマイクロアレーを用いて網羅的に比較するため予備検討として、ヒト糸球体とヒト腎皮質、髄質との比較を行い、それに特異的に発現する遺伝子を明らかにした。 3)正常糸球体タンパク質を二次元電気泳動し、ゲルを鍍銀染色で検出した約1000スポットをMALD-TOT質量分析機で分析し、約500スポットのタンパク質を同定した。その結果にタンパク質の性格や機能などをつけたデータベースにした。 4)マイクロアレーでヒトIgA腎症腎組織で発現が亢進している遺伝子として、新規遺伝子を分離した。この遺伝子から翻訳されるタンパク質はムチンファミリーに属した膜タンパク質であり、近位尿細管に発現していた。この分子は増殖因子の受容体に結合し、増殖因子の機能を抑制する分子であった。 5)ヒトIgA腎症糸球体をLaser-capture microdisection法で糸球体を分離して、その遺伝子発現、タンパク質の変化の検出を試みているが、サンプル量が少なく、通常の二次元電気泳動での解析では、ヒトIgA腎症の病態特異的タンパク質は検出できなかった。
