肥満遺伝子産物レプチンの糖脂質代謝亢進作用の分子機構の解明

研究概要	脂肪萎縮性糖尿病では体脂肪量の減少に伴って低レプチン血症、インスリン抵抗性糖尿病、高脂血症および脂肪肝が認められる。我々は既にレプチン過剰発現トランスジェニックマウス(LepTg/+)と脂肪萎縮性糖尿病マウス(A-ZIPTg/+)の交配実験により、A-ZIPTg/+マウスにおける糖脂質代謝異常が改善することを報告した。今回、脂肪萎縮性糖尿病におけるレプチンの糖脂質代謝改善作用の分子機構を明らかにするために、LepTg/+とA-ZIPTg/+を交配することにより得られる15週齢雄性F1マウス(+/+、LepTg/+、A-ZIPTg/+、LepTg/A-ZIPTg)およびレプチン投与24時間後のA-ZIPTg/+マウスの肝臓における遺伝子発現プロフィールを検討した。Affimetrix社製のマイクロアレイを用いた検討によりAZIPTg/+マウスでは+/+マウスに比較して解糖系、ペントースリン酸回路および脂肪酸合成及び分泌に関与する遺伝子の発現亢進が認められた。これに対して糖新生および脂肪酸酸化に関与する遺伝子の発現は低下していた。一方、LepTg/A-ZIPTgマウスではA-ZIPTg/+マウスで認められる遺伝子発現の変化が+/+マウスのレベルにまで改善していた。レプチンを投与したA-ZIPTg/+マウスでは解糖系、ペントースリン酸回路および脂肪酸合成及び分泌に関与する遺伝子の発現は正常化したものの、脂肪酸酸化に関与する遺伝子発現の低下は回復しなかった。以上より、脂肪萎縮性糖尿病マウスにおいてレプチンによる高中性脂肪血症および脂肪肝の改善作用は高インスリン血症等の改善による二次的な作用であると考えられた。
