サルES細胞からの肝細胞分化誘導および増殖に関する基礎的研究

研究概要

○目的
1.サルES細胞を培養し、肝実質細胞への分化誘導を確認する。
2.分化の確認された細胞を分離培養する。
3.肝細胞としての機能評価と人工肝への応用(in vitro, in vivo)の基礎を確立する。
これらの目的を達成するためにまず扱いの容易なマウスES細胞の肝細胞への分化誘導に着手した。続いてサルES細胞のin vitroでの肝細胞への分化誘導に着手し、nude mouse肝障害モデルを用いてin vivoでの肝細胞への分化誘導に取り組んだ。同時に将来の細胞移植療法を視野に入れ、MRIを用いた移植細胞のトラッキング法を確立した。

○現在までの研究進行状況
1.マウスES細胞に各種成長因子を投与し、in vitroで肝細胞への分化誘導を行ってAlb、AFPなどの肝細胞特有な遺伝子の発現を確認した。
2.マウス肝細胞とマウスES細胞との共培養法による肝細胞への分化誘導法を行い、培養開始後1週間でAFP、Albの発現を確認した。
3.サルES細胞にaFGFを投与し、in vitroで肝細胞への分化誘導を行い、Albなどの肝細胞特有な遺伝子の発現を確認した。さらにヌードマウス肝障害モデルに移植し、肝での移植細胞の増殖とアルブミンの発現を確認した。
4.分化誘導した細胞を移植する際、無侵襲下に細胞をトレースする新しい方法(MRIを用いた細胞追跡法)を確立した。HGF、FGF、EGFで肝細胞へ分化誘導したマウスES細胞を鉄にて標識した後、ヌードマウス肝障害モデルに移植し、無侵襲下に経時的にMRIで移植細胞を追跡した。
