消化器癌の新規遺伝子増幅領域の網羅的探索と癌関連遺伝子の同定

研究概要	新規増幅領域から消化器癌の発症に関与する遺伝子を同定した。各種腫瘍においてゲノムコピー数異常の網羅的解析を行い、見出されてきた新規の遺伝子増幅領域を標的に位置候補遺伝子アプローチで癌関連遺伝子の単離を進めた。CGH法によりゲノムコピー数異常を探索し、総計860例の解析を終えた。そのうち、消化器癌は、食道扁平上皮癌(細胞株42、症例29)、胃癌(細胞株25、症例105)、肝細胞癌(細胞株11、症例70)、膵癌(細胞株22)、大腸癌(細胞株5)、ならびに薬剤耐性関連腫瘍(細胞株22)である。その他の細胞株(22)の解析を終了した。食道扁平上皮癌(ESC)における新規11q22増幅の標的がアポトーシス阻害cIAP1であることを明らかにした。肝細胞癌(HCC)のゲノム異常スクリーニングの結果、13q34と8qの増幅を高頻度に検出した。詳細な解析から13q34増幅の標的が転写因子DP1に加え、ユビキチン・リガーゼE3め構成分子CUL4A(cullin4A)、ユビキチン・リガーゼAPCのサブユニットCDC16(cell division cycle 16)であると同定した。HCCにおいてはDP1は腫瘍径と明らかな相関があり、その機能亢進は悪性化に直接関与することも明らかにした。加えて、腫瘍ゲノムコピー数の網羅的・高精度解析をめざし、高密度CGHアレイを開発し、すでに800個の癌関連遺伝子を張り付けた「癌の個性診断」用high through-put CGHアレイを実用化した。
