MAGEファミリーに属するグリオーマ特異的な新規遺伝子の臨床応用と機能解析

研究概要	MAGE-E1a cDNAの塩基配列から予想される蛋白のアミノ酸配列のうち、MAGE-conserved domainを含まないようにしてN末端側、C末端側に14merのペプチドを作成した。これをウサギに免疫してペプチド抗体を作成した。これらの抗体はともに58kDaの単一のバンドを認識した。RT-PCRでMAGE-E1遺伝子が発現していないことが確認できた正常肺組織ではこのバンドは認められなかった。以上の結果より、この58kDaのバンドがMAGE-E1蛋白であると判断した。U87MG細胞より抽出した蛋白分画を用いたウェスタンブロット解析や同細胞を用いた免疫染色の結果、MAGE-E1蛋白は細胞質、特に核の周辺に局在していることが明らかとなった。MAGE-E1とGFPの癒合蛋白をHIH3T3,HEK293細胞に強制発現させて機能解析を試みた。MAGE-E1遺伝子を強制発現させると細胞が死滅したため、MAGE-E1の過剰な発現は細胞障害性があると考えられた。MAGE-E1が悪性グリオーマの増殖に関わっていることを示すために以下の実験を行った。T98G細胞を無血清培地で72時間培養することによって細胞をG0期に入るようにすると、通常の10%FBSで培養したもとと比較してMAGE-E1蛋白の発現は減少した。これらの細胞の免疫染色の結果、蛋白レベルの減少は特定の細胞に起因するものではなく、すべての細胞で一様に減少していた。グリオーマの摘出標本を用いたウェスタンブロット解析の結果、膠芽腫では星細胞腫に比べて蛋白レベルが高かった。また、免疫染色の結果も膠芽腫では蛋白発現が高い細胞がびまん性に存在していたが、星細胞腫では発現が比較的高い細胞が散在していた。以上の結果より、MAGE-E1蛋白は悪性グリオーマで発現が上昇しており、グリオーマ細胞の増殖に関与していることが示唆された。
