研究概要

膀胱癌細胞株において、マイクロアレイ解析で親株とシスプラチン耐性株との間に発現の差を認める遺伝子の中からInositol 1,4,5-triphosphate (IP_3) receptor type1 (Ip_3R1)遺伝子を同定したIP_3R1は小胞体膜上に位置し細胞質内Ca濃度を調節するとともにアポトーシスを誘導する。シスプラチン耐性株T24/DDP10では、IP_3R1を強発現させシスプラチン刺激を加えると、アポトーシスが誘導された。細胞内Ca濃度の上昇のみでもアポトーシスは誘導された。またsi-RNAの手法によりIP_3R1を発現抑制するとアポトーシスを抑制し、Caのキレート剤にてもアポトーシスを抑制した。これらの結果より細胞内Ca濃度を調節するIP_3R1はアポトーシスの誘導に関連していることが示唆された。またMTSアッセイでは、IP_3R1の発現とCa濃度の上昇により耐性株のシスプラチンに対する感受性を上昇させ、IP_3R1の発現抑制とCa濃度の低下により親株のシスプラチンに対する感受性を低下させた。以上より膀胱癌のシスプラチン耐性獲得のメカニズムはIP_3R1の発現抑制により、アポトーシスに対し抵抗性になることによる可能性が示唆された。シスプラチンによる加療を行った臨床検体における治療前後のIP_3R1の発現パターンを解析した。結果をもとに、将来的には抗癌剤耐性化予測因子としての活用や、IP_3R1の強発現またはCaによるシスプラチン耐性癌の治療への応用を考えている。
