研究概要	抗真菌剤を指向したスフィンゴ脂質代謝阻害剤の研究:カフレファギン(KF)は、真菌特異的なスフィンゴ脂質・イノシトールホスホセラミドの合成酵素阻害剤であり、抗真菌活性を有する。化学合成したKFおよびその誘導体を用い、活性構造相関を解析した。KFの有する糖部分の水酸基の立体構造が抗真菌活性に重要であることなどが明らかとなった。 LPS受容体の異物識別機構に関する研究:内毒素の本体であるLPSが宿主に認識されるために必要な宿主因子としてToll-like receptor (TLR) 4とMD-2が必要であることが示されている。そして両者は細胞表面上で複合体を形成している。そこで、TLR4と細胞表面上で複合体を形成するために重要なマウスMD-2中のアミノ酸残基と異物認識に重要なマウスMD-2中のアミノ酸残基をアラニンスキャン法により決定した。アミノ含有脂質によるマクロファージ活性化に関する研究:アミノ含有脂質であるSer-Gly含有脂質にはマウスマクロファージを活性化してTNFαの産生を誘導する活性がある。細胞発現系を利用してSer-Gly含有脂質認識に関わる細胞膜蛋白質を解析した。その結果、Ser-Gly含有脂質認識にはTLR4とMD-2両方が必要であることが明らかになった。
