生物活性二次代謝産物に見る多様性の分子基盤の解析と展開

研究概要

本研究は、二次代謝系の多様性発現基盤を遺伝子解析と反応特異性の観点から明らかにし、有用物質生産に資することを目的とし、(1)ラクタムポリケチド ビセニスタチンの生合成における糖転移酵素遺伝子vinCの破壊株を樹立し、またスタータ活性化に関わるvinM及びvinNの発現系を確立し、酵素精製と機能解析を進めた。(2)VinCを大量精製し、基質本体をdTDP-ビセニサミンと証明した。さらにアグリコンの異性体ネオビセニスタチンアグリコンを新たに合成し、糖転移反応が天然型と同効率で進行することを示した。全く異なる骨格を持つBrefeldin A及びEstradiolをビセニサミン受容体として全く新規な配糖体の創製に成功し、VinCが適切な疎水性結合と分子サイズにより多様なアグリコンを認識出来ることを示した。(3)ビセニスタチン生産菌に発見した新規ポリケチド合成酵素に対応する代謝物を精査し、新規28員環マクロリドHalstoctacosanolide A及びBを単離構造決定し、その生合成遺伝子の約70%を解読した。(4)イソプレノイド生合成の精密解析プローブとして完全重水素化メバロノラクトンの実用的不斉合成法を開発し、放線菌のジテルペン環化酵素の反応機構を動的に解明した。(5)放線菌Streptoalloteichus hindstanusのアミノ配糖体生合成遺伝子の解析を進め、糖質環化酵素遺伝子を大量発現して機能を確認した。クラスター内での読み枠の配列はブチロシンのそれと相違し、糖質環化酵素とアミノ基転移酵素が隣接していた。放線菌に比し、Bacillus属の遺伝子クラスターの特異性が明らかとなった。(6)ブチロシン生合成においてパロマミン生成に関わるグルコサミン活性化酵素BtrD及び糖転移酵素BtrMについて、破壊株の造成または発現酵素による反応解析により機能を明らかにした。
