研究概要

1.アスパラギン結合型糖鎖と細胞内レクチン/分子シャペロンとの相互作用による新生タンパク質の品質管理に関して、我々は昨年度小胞体内シャペロンであるカルネキシン(CNX)及びカルレティキュリン(CRT)のリガンドと考えられている12糖(Glc_1Man_9GlcNAc_2)の合成を達成しNMRによるCRTとの相互作用の観測にも併せて成功した。今年度は等温滴定型カロリメトリー(ITC)による様々な糖鎖とCRTの結合の定量解析を行なった。また不良糖タンパク質の分解に係わると考えられているマンノシダーゼ様タンパク質EDEMのリガンド候補として有力な10糖Man_8GlcNAc_2及び11糖Glc_1Man_8GlcNAc_2の合成を達成した。更に、合成糖鎖をタンパク質に組み込むための手法を種々検討した。
2.C-マンノシルトリプトファンの生体機能の解明とタンパク質構造に与える影響を調べるため、これを含むタンパク質の様々な部分構造を合成した。
3.可溶性高分子担体を用いる糖鎖合成において、生成物を簡便に釣り上げて純粋な形で単離する手法を開発した。これを応用し、N-アセチルグルコサミン転移酵素-Vの阻害剤としてデザインした2基質型化合物の合成ルートを開発した。
4.シアル酸は複合型糖タンパク質糖鎖や糖脂質に広く存在する重要な糖である。本年度はシアル酸供与体のエステル置換基を変換する手法を開発し、グルコシル化反応における置換基の影響を系統的に調べた。
5.CMP-シアル酸とN-アセチルラクトサミンを同一分子内に組み込んだ2基質型のシアル酸転移酵素阻害剤をデザインし、合成した。2つのコンポーネントを結ぶリンカーの長さを系統的に変化させ阻害活性との相関を検討した。その結果、比較的低濃度でα2,3及びα2,6-シアル酸転移酵素を阻害する化合物を見い出した。
