止血血栓に関与する蛋白質とその結合蛋白質複合体の結晶構造解析

研究概要	プロトロンビンやX因子の迅速な活性化反応は、カルシウム及びボスファチジルセリン依存性の生体膜上のリン脂質急速なセリンプロテアーゼの前駆体活性化反応として重要である。しかし、凝固因子の生体膜リン脂質上での活性化や分子レベルの結合機構や酵素反応の増幅メカニズムについての分子レベルの解析は殆ど行われていない。我々が単離したヘビ毒由来の抗凝固タンパク質は、Glaドメインとモル比1:1で強固に結合し抗凝固活性を示すことを明らかにしてきた。従って、これらのGlaドメイン結合タンパク質はGlaドメインの機能を解析する有力なツールである。今回、凝固IX因子のリン脂質結合部位であるGlaドメインとIX-bpの複合体の結晶化とその結晶構造解析を行ない、血液凝固因子のGlaドメインとリン脂質の結合様式を分子レベルで解明した。IX-bp(凝固IX因子結合タンパク質)をハブ毒1gから5mg単離した。これを7回繰り返し、約35mg単離した。一方、ウシ血漿25lから凝固IX因子を5mg単離した。これを5回繰り返し、IX因子を25mg単離した。IX因子のAsp-Nプロテアーゼによる限定分解によりIX因子のGlaドメインを断片化後、Glaドメインペプチド(残基1-46)を3mg単離した。IX因子GlaドメインペプチドとIX-bpを等モル混合し、1mM Caと5mM Mg存在下でのゲルろ過により複合体を単離した。結晶化の条件は5mM Ca^<2+>,2mM Mg^<2+>,7%PEG6000,pH8.0と決定した。構造解析:水野洋博士との共同研究で決定した構造既知のIX/X-bp(Nature Str.Biol.で発表)を利用した分子置換法によりGlaドメイン複合体の立体構造を決定した。
