tRNA擬態分子による翻訳終結機能の分子機構解明

研究概要

tRNA擬態性蛋白質について、昨年までの研究成果を発展させることで新たに以下の知見を得た。
原核生物ペプチド鎖解離因子上に、昨年までに同定したペプチドアンチコドン機能制御領域である"ドメインC"は、近年明らかにされたペプチド鎖解離因子の立体構造上、実際に、ペプチドアンチコドンに近接するドメインであることが明らかになった。既に公表した機能変異部位が、このドメインの特定の側面部位に局在することから、この領域がリボソーム上でペプチドアンチコドン部位を定位させることで、コドン認識・識別を正確に行うという機能モデルを公表した。
真核生物ペプチド鎖解離因子のコドン識別機能ドメインを特定する為に、既知の立体構造情報を応用し、通常の3つの終止コドン(UAA, UAG, UGA)を認識する出芽酵母由来eRF1と、例外的な終止コドン(UGAのみ)を用いるテトラヒメナ由来のeRF1とのドメインごとのキメラ体産生遺伝子をデザインした。このキメラ体蛋白質および、同蛋白質に、系統的な変異導入を行った変異体のin vivoおよび、in vitro解析結果により、ドメインIがコドン特異性を決定することを明らかにした。
また、翻訳終結過程の最終段階であるリボソーム再生過程における、伸長因子EF-Gと、リボソーム再生因子との相互作用ドメインを系統的な遺伝学手法と、生化学手法により検索した。その結果、リボソーム再生因子との相互作用部位として、EF-GのtRNA擬態領域であるドメインIV-Vの立体構造上の特定の領域に局在するアミノ酸残基群を特定した。
以上の、研究成果により、「解離因子-tRNA擬態仮説」の具体的な実証を進めつつ、さらに、翻訳終結に関わるその他の蛋白質群が、tRNA擬態性ドメインを有効な機能領域として用いている事を明らかにできた。
