高等植物の生物時計とサーカディアンリズムの統御ネットワークの分子基盤

研究概要

高等植物における生物時計機構は、極めて重要な植物高次制御機構の一つである。植物の成長、花の開花時期、気孔の開閉、胚軸の伸長など、実に様々な機能が概日リズム(時計)により支配されていると考えられている。しかし、植物の時計とは如何なる物か、時を刻む生物的分子機構とはどのようなメカニズムかに関する研究は漸く始まったばかりである。本研究計画において我々は、シロイヌナズナをモデル植物として、概日リズムを刻む生物時計の分子実体とそのメカニズムに関する解析を進めてきた。その結果、生物時計と密接に関連して機能すると思われる5つのファミリー遺伝子群(APRR1〜APRR9)を発見した。これに関した解析から、次のことを明らかにした。
(1)これら5つに遺伝子は連続的で規則的なリズムを刻む。
(2)APRR9は光誘導を受ける。
(3)APRR1の過剰発現植物には時計機能が大きく影響される。
(4)APRR9の過剰発現植物では開花時期が顕著に早まる。
(5)APRR5の過剰発現植物では光に対する感受性が高まり、光形態形成に顕著な以上が見られる。
以上の結果から、我々が発見した時計関連遺伝子群は、単に時計機能のみならず、開花時期の制御、光形態形成制御といった高等植物の高次機能制御の本質に関わる重要な機能を果たしていることが明らかとなった。これらの、結果は従来まで考えられていた時計分子機構に全く新しい視点を付け加える点で極めて興味深い。
