ホヤ卵細胞質内で局在する組織決定因子の同定と解析

研究概要

ホヤは典型的なモザイク卵であり、発生の早い時期から各細胞は自律的分化能を持つ。その原因として、卵細胞質内組織決定因子が卵の中で既に特定の時期に片寄って存在していることが知られている。マボヤでは、受精卵の時期から植物半球後方の卵細胞質(PVC)に片寄って存在する母性因子が、筋肉の自律的分化、間充織・脊索の反応能に加え、胚後極の卵割パターン、前後軸形成などの過程において重要な役割を果たすことが、PVCの除去・移植実験などから示唆されている。現在までに、cDNAプロジェクトなどによって、受精卵の時期からPVCに片寄って存在するmRNAが多く発見され、それらはTypc1 postplasmic mRNAと呼ばれている。マボヤのTypc1 mRNAには、macho-1、HrPEMなどが既に知られており、解析がなされている。我々は、マボヤ卵のPVCに局在する母性mRNAの機能解析を目的として、マボヤ卵に存在する母性mRNAのcDNAプロジェクトデータベース(MAGEST)のデータから、8細胞期の胚後極に局在するmRNAを網羅的に選び出し、受精卵、8細胞期においてin situ hybridizationを行った。その結果、多くのクローンは受精卵の時期には局在しないが8細胞期までに胚後極に局在するTypc2 mRNAであることが明らかになった。Type1 mRNAは、既知のものも含めて9種類であった。そこで、Type1 mRNAの中で、全長の塩基配列がわかっていなかったものについてその配列を明らかにし、いくつかは脊椎動物の遺伝子とホモロジーが高いことがわかった。それらは、macho-1、HrPEM、RNA結合タンパク(HrPEM-3)・Hrwnt-5、Zn fingerタンパク(HrZF-1)、serine/threonine kinase(HrPOPK-1)、glucose transporter、g1-related protein、no homologyの9種であった。現在、それらの初期発生における発現パターンを詳細に調べるとともに、6つのType1遺伝子について、アンチセンスモルフォリノを用いた機能解析を行っている。
