ホヤ卵細胞質内で局在する組織決定因子の同定と解析

研究概要

多くの動物の卵では、卵内情報は初期発生に関して重要な働きを持っている。しかし、脊索動物胚での母性因子の機能は十分に理解されていない。マボヤでは、植物半球後方の卵細胞質(PVC)に片寄って存在する母性因子は前後軸形成、すなわち、筋肉・間充織の形成、生殖細胞形成、連続した不等分裂・特定のmRNAが胚後極の構造(Centrosome-Attracting Body:CAB)へ濃縮される、細胞周期の遅れ等の過程において重要な役割を果たすことが、PVCの除去・移植実験などの顕微胚操作実験から示唆されている。
現在までに、受精卵の時期からPVCに片寄って存在する9種類のmRNAを同定し、その全長配列を決定した。これらの発見飽和度から、マボヤの卵内に存在するType I postplasmic RNAはこの9種類でほぼ網羅されていると考えられる。次に、特異的な翻訳阻害剤として働くモルフォリノアンチセンスオリゴ(M-oligo)を用い、今までに機能解析が行われていなかった7つのtype I、遺伝子(HrPEM、HrPOPK-1、HrWnt-5、HrGLUT、HrPEM-3、HrPEN-1、HrPEN-2)について、網羅的な機能阻害実験を行って組織分化における影響について調べた。その結果、type I mRNAは初期胚発生において、様々な働きを担っていることが示唆された。特にHrPEM(相同性なし)が連続した不等分裂に関わるらしいこと、HrPOPK-1(カイネース)が特定のmRNAのCABへの濃縮に関与していることがわかりつつある。これらの現象はともに興味深く、今後、詳細な解析の価値がある。
