研究概要

広義の大脳皮質である海馬と嗅球を構成するニューロン群、主に、GABAニューロンサブポピレーションの定量解析及びニューロン相互関係の解析を進めた。マウス海馬においてGABAニューロン群の特徴あるサブポピュレーションであるカルシウム結合蛋白parvalbumin(PV)、calbindinD28k、calretinin含有ニューロンの領域、層別の分布蜜度についてステレオロジーによる定量解析を進めた。また、海馬アストロサイトの構造解析,およびoptical disector法による定量解析も行った.ゴルジ染色標本の超高圧電子顕微鏡による立体観察では個々のアストロサイトが細かい突起からなるmassを形成することが示された.アストロサイト同士の関係を明らかにするために,二つの隣接するアストロサイトに細胞内蛍光色素注入を行い共焦点レーザー顕微鏡観察を行ったところ,重なりは限定されており個々のアストロサイトは異なった領域(ドメイン)を占めていることが明らかとなった.optical disectorを用いた定量解析ではS100免疫反応性細胞として同定されるアストロサイトが層や領域間で大きな違いがなく比較的一定した密度を持ち,個々の細胞がドメイン構造をとることが示唆された。
一方、嗅球においては、機能単位である糸球体のニューロン相互のシナプス結合を解析した。嗅入力を直接受ける介在ニューロンと受けないグループ(それぞれタイプ1、タイプ2傍糸球体細胞と名づけた)それぞれがフィードフォワード、フィードバック的に働くことを示唆した。また、糸球体における投射ニューロン樹状突起上に嗅神経とタイプ2介在ニューロン突起とがモザイク状に分布しシナプス結合していること、嗅神経終末は決して従来いわれているように細い樹状突起先端部にのみ分布しているのではなく、太い近位部にもシナプス結合していた。
