大脳皮質の構成要素・機能素子の比較解剖学的、形態学的定量解析・三次元構造解析

研究概要

広義の大脳皮質である海馬と嗅球を構成するニューロン群、主に、GABAニューロンサブポピレーションの定量解析及びニューロン相互関係の解析を進めた。マウス海馬において前年度のGABAニューロン群サブポピュレーションであるカルシウム結合蛋白parvalbumin (PV)、calbindin D28k、calretinin含有ニューロンの定量解析に引き続き、別のGABAニューロンサブポピュレーション群、ペプチド含有GABAニューロンの定量解析を進めた。CCK, VIP, somatostatin, neuropeptide Y含有ニューロンについて、optical disector法による定量解析を進め、完了した。これらの解析により、マウス海馬においてGABAニューロン群の構成がほぼ明らかになり、これまでのラットでの所見との比較が可能になった。また、ペプチドであるCCKの海馬での存在様式、特に、主ニューロンでの存在がラットとマウスで大きく異なることが明らかとなり、マウス海馬での背腹方向での差異等その解析を進めた。
一方、嗅球においては、そのニューロン・シナプス構成をいくつかの動物種で検討し、我々が提唱している1型、2型の2種類の傍糸球体細胞が普遍的に存在すること、これまで食虫目であるジャコウネズミにしか存在が示されていなかった特異なシナプス野nidusが他の食虫目であるモグラにも存在するが、別の食虫目ハリネズミや、その他には存在しないことを明らかにした。
