コンディショナルターゲッティングを用いた脳高次機能の分子解析

研究概要	本研究の目的は、シナプス形成、維持、精緻化などに関与が示唆されてきたシナプス機能分子群がシナプス可塑性発現と、記憶・学習を始めとした脳高次機能に果たす役割を明らかにすることである。上記の目的を達成するために、我々が開発を進めてきたCre-loxP組換え系を用いたコンディショナルノックアウトマウスのシステムを用いて研究をおこなった。まず、単純なノックアウトでは致死であり、シナプス結合の精緻化に関与することが示唆されているNMDA型グルタミン酸受容体ε2サブユットに着目し、この遺伝子にloxP配列を組み込んだ標的マウスε2loxPを樹立した。一方、記憶の形成や学習に関与すると考えられる海馬で部位特異的にCre組換え酵素による遺伝子組換えを起こすために、海馬CA3錐体細胞に優位に発現するカイニン酸受容体γ1サブユニット遺伝子に着目し、このプロモーター制御下にCreを発現するγ1CreNマウスをC57BL/6のES細胞より作成した。このマウスの遺伝子組換え能を調べるために、Cre活性によりlacZが発現するCAG-CAT-Z11トランスジェニックマウスと交配し、Cre発現部位、組換え効率を同定した。その結果、海馬CA3錐体細胞で効率よく組換えが誘導されることが明らかになった。そこで、このγ1CreNマウスとε2loxPマウスを交配させ、loxP配列遺伝子をホモに持ちCre遺伝子をヘテロに持つ固体を選別した。このマウスのε2サブユニットの発現を免疫組織科学的に調べた結果、海馬CA3領域で選択的にε2サブユニットが欠損していることが明らかになった。現在このマウスとの表現型を形態学的、電気生理学的および行動解析学的手法により解析を進めており、シナプス機能タンパクと脳高次機能の関連を明らかにしていく。
