液胞型H^+-輸送性ピロホスファターゼを対象とした抗マラリア薬の探索と展開研究

研究概要

液胞型H^+輸送性ピロホスファターゼ(H^+-Ppase)は、植物、光合成細菌の他に、マラリア病原虫を含む原生動物にも存在する。マラリア病原虫ではacidocalcisomeと呼ばれるオルガネラ膜に局在する。動物には存在しない利点を考慮し、寄生病原虫に対する薬剤開発のターゲットと考えた。本年度は、カイメン、サンゴ、紅藻、ホヤなど計239種の海洋性生物由来の抽出液を出発物質として、H^+-Ppase阻害剤を検索した。その結果、とくに阻害活性の高い物質として、5種のアシルスペルミジンを同定した。精製阻害物質は、酵素:阻害物質=1:35で完全に阻害した。これらのうち3種は未知物質であった。いずれもH^+-Ppaseの加水分解活性を阻害するのみでなく、H^+輸送活性も強く阻害する。今後、抗マラリア薬としての有効性を検証するためには、阻害の特異性についての検討が必要である。また、有機化合物以外に、金属イオンによる阻害作用についても検討した。ニッケル、マンガン、コバルト、銅、鉛、2価鉄、カドニウム、ストロンチウムは0.1mMの濃度で強く阻害することを見出した。いずれもMg^<2+>を必須とするH^+-PpaseのMg^<2+>と競合するものと思われる。
