液胞型H^+-ピロホスファターゼを対象とした抗マラリア薬の探索と展開研究

研究概要

マラリアは、マラリア病原虫プラスモディウム属がハマダラカを媒体として伝染し、ヒト肝細胞・赤血球に寄生することによる疾患である。本研究は、マラリア原虫がもつH^+輸送性ピロホスファターゼ(H^+-PPase)に焦点をあて、この酵素に対する特異的阻害剤の探索を通して、抗マラリア特効薬を見出し開発することを目的とした。
1.阻害剤に関する研究成果(新規阻害剤の発見)
沖縄の海洋に生息する生物のうち,軟サンゴから得られた成分がもっとも強い阻害活性示した。その成分の化学構造を決定したところ分岐したアシル基をもつアシルスペルミジン類縁化合物であることが判明した。基質加水分解・プロトン輸送活性を強く阻害した(50%阻害濃度1μM)。さらに生きた植物細胞においてもH^+-PPaseを阻害し,その生理機能を強く抑制することが証明された。
2.H^+-PPaseに関する研究成果・新知見
(1)植物H^+-PPaseの遺伝子破壊株の解析により遺伝子欠損は生育の著しい抑制をもたらすこと,ずなわち本酵素が植物体の正常な生育に不可欠であることを明らかにした。
(2)ヤエナリH^+-PPaseの構造・機能協関の解析により,少なくとも2つの細胞質側親水性ループが基質結合・触媒部位を形成し,その中の保存性の高いアミノ酸残基が基質加水分解を司っていることを明らかにした。
(3)変異導入とそれに引き続く機能検定のやりやすい大腸菌発現系の確立を目的に,放線菌H^+-PPaseを対象に解析を進めた。実験系の確立に成功し,放線菌H^+-PPaseの固有の性質を明らかにした。
(4)H^+-PPase機能の直接測定のためのパッチクランプ法を世界に先駆けて開発し,H^+-PPaseの特質,分子活性をもっとも精度の高い方法で明らかにした。
