日本産魚類のミトコンドリアゲノム・データベースの構築

研究概要	水域生態系の重要構成要素である魚類は,資源生物としてもきわめて重要なグループである。この魚類には,多様な系統に属する2万5千もの種が存在しており,卵や稚仔魚を含むあらゆるタイプの試料の種の正確な識別と分類,遺伝的多様性の把握,種内集団(系群)の峻別,分子系統分析などに,各種のDNA塩基配列情報が重要かつ不可欠になりつつあり,急速に増大するDNA情報を使いやすい形で整理・蓄積することが強く求められている。そこで本研究では,種々の面から有用性の高い情報源であることが明らかになっているミトコンドリアゲノムを取り上げ,「魚類のミトコンドリアゲノム・データベース」を構築することを目指した。プロジェクト3年間で,1)国内および国外からさまざまな系統を代表する約数百種の貴重な標本試料を新たに入手し,2)確立したロングPCRと魚類ユニバーサルプライマーを活用する方法を用いて約200種のミトコンドリアゲノムの全塩基配列を決定した。3)加えて,同ゲノムの部分塩基配列を約400種について得た。4)魚体標本の多くは,国立科学博物館ないし千葉県中央博物館へ登録した。そして5)最終目標である「魚類のミトコンドリアゲノム・データベース」を完成させ,インターネット上に公開した。URLはhttp://mitofish.ori.u-tokyo.ac.jp/であり,ミラーサイトが国立科学物館にある。本データベースは国際的使用が可能な英語仕様である。機能としては,国際的DNAデータベースにミトコンドリアゲノムの全塩基配列データが登録されているすべての魚類の情報が検索できる。また,このデータベースに対して,任意の塩基配列の相同性検索ができるので,種や魚類グループの識別に有用である。今後、さらに充実を図る予定であるが,データの増大に伴ってその有用性は飛躍的に大きくなるはずである。
