研究概要	目的 HTLV-I taxはHTLV-I感染によるシェーグレン症候群(Sjogren's syndrome : SS)発症の中心的役割をはたしている。HTLV-I感染によるSS発症機序を検討するために、HTLV-I taxの抗アポトーシス分子機序を検討した。方法 HTLV-I tax遺伝子導入細胞であるJPX-9細胞を用い、種々のアポトーシス刺激を加え、HTLV-I taxの抗アポトーシス分子機序をDNA断片化、caspase活性化、ミトコンドリア膜電位低下、Bcl-2関連蛋白発現の検討で評価した。結果 Taxが発現したJPX-9細胞は、受容体依存性アポトーシス刺激(抗Fas抗体、TRAIL)および受容体非依存性アポトーシス刺激(PDTC、etoposide、staurosporine)に対して抵抗性を示し、caspase活性化とDNA断片化はHTLV-I tax発現により有意に抑制された。HTLV-I taxのアポトーシス抵抗性はミトコンドリア依存性に誘導され、上述のアポトーシス刺激におけるミトコンドリア膜電位の低下は抑制された。しかしながらHTLV-I tax発現の有無でのBcl-2、Bcl-xL、Bax発現に差異は認めなかった。考察 HTLV-I taxはT細胞の活性化誘導型細胞死を抑制し、この事象はサイトカインや自己抗体の産生を促すことでSSの発症に関与すると考えられた。HTLV-I taxはミトコンドリアに作用するが、今回の検討では標的分子を同定することはできなかった。今後はcDNA arrayでの標的分子のin vitroでの同定やSS症例血球細胞を用いての評価を行い、HTLV-I感染SSの発症に関与する遺伝子群の同定を試みる。
