研究概要	デスモコリン1-3(Dsc1-3)の有核細胞発現クローンを用いて、PCR法で細胞外部位のcDNAを増幅してバキュロウイルスに組込み、昆虫細胞にトランスフェクトしてバキュロ蛋白を作成する。His-tagを利用してニッケルカラムによりバキュロ蛋白精製し、高次構造エピトープを壊さないリコンビナント蛋白の作成に成功した。この蛋白を用いて、Dsc1-3IgG/IgA抗体を検出するELISA法を確立した。また、BP230、エンボプラキン、ペリプラキンのそれぞれについて、分子を3部位に分けて、全長をカバーするのcDNAを、ケラチノサイトcDNAライブラリーをテンプレートとしたPCR法で単離、大腸菌発現ベクターpGEXにクローニングし、リコンビナント蛋白を作成した。それを用いた免疫ブロット法を確立し、水泡性天疱瘡、腫瘍随伴性天疱瘡の自己抗体を検出するとともに、その他の疾患におけるこれらの抗原に対する抗体を検出した。さらに、培養ケラチノサイトの上清を大量に回収し、硫安塩析で濃縮し、さらに各種カラム精製法を用いて、ラミニン5、120kD/97kD LAD1(透明層型線状IgA水庖性皮膚症抗原)を単離した。一方、これらの精製抗原を用いた免疫ブロット法で、透明層型線状IgA水庖性皮膚症と抗エピリグリン瘢痕性類天庖瘡抗原のラジオアイソトープを用いない診断法を確立した。また、腫蕩随伴性天庖瘡の診断のためにラジオアイソトープ標識細胞を用いた免疫沈降法が必須であるが、ラジオアイソトープを使用しない免疫沈降法・免疫ブロット法コンビネーション法を検討中である。現在、全く未知であるIEN型のIgA天庖瘡抗原およびVII型コラーゲンに反応しない基底板下部型線状IgA水庖性皮膚症の抗原を同定するための基礎的実験をはじめた。
