研究概要	まず、pET大腸菌発現系を用いて詳細なBP180のリコンビナント蛋白を作成し、その蛋白を用いた免疫ブロット法を確立し、瘢痕性類天疱瘡がBP180の最もC末端の100アミノ酸に反応することを証明した。また、in vitro traslation発現系を用いてBP230のリコンビナント蛋白を作成し、その蛋白を用いた免疫ブロット法で多くの水疱性類天疱瘡血清がBP230のC末端部に反応することを示した。また、線維芽細胞発現系を用いてデスモコリン1-3 (Dsc1-3)のトランスフェクション法を改良、IgA天疱瘡診断の診断手段として利用であることを示した。さらに、BP230、エンボプラキン、ペリプラキンのそれぞれについて、分子をおよそ3倍位に分けて、全長をカバーするのcDNAを、ケラチノサイトcDNAライブラリーをテンプレートとしたPCR法で単離、大腸菌発現ベクターpGEXにクローニングし、リコンビナント蛋白を作成した。それを用いた免疫ブロット法を確立し、水疱性類天疱瘡、腫瘍随伴性天疱瘡の自己抗体を検出するとともに、その他の疾患におけるこれらの抗原に対する抗体を検出した。現在まで、抗ラミニン5瘢痕性類天疱瘡の診断のためにラジオアイソトープ標識細胞を用いた免疫沈降法が必須であったが、私共にラジオアイソトープを使用しない方法として、精製免疫ラミニン5を抗原とした免疫ブロット法を確立し、抗ラミニン5瘢痕性類天疱瘡の診断に有用であることを示した。また、現在、線状IgA水疱性皮膚症(LAD)の抗原として、表皮から抽出される79kDa LAD1抗原が知られているが、この抗原の抽出には多量の皮膚を要する。私共は、より簡便な方法として、培養ケラチノサイトの培養上清を濃縮したサンプルを抗原とした免疫ブロット法を確立し、多くのLAD血清が120kDa LAD1に反応することを示し、LADの診断に有用であることを示した。
