研究概要	1 インフルエンザウイルスの宿主への吸着および宿主からの発芽の両者を阻止する新規シアロ化合物の発見:シアル酸の3位にフッ素を導入したシアリルホスファチジルエタノルアミンは、多くのインフルエンザウイルスヘマグルチニンおよびノイラミニダーゼ両者の機能を二重阻止できることを発見した。このように感染の初期および終期の両方をダブルブロックできる物質は、これが最初の報告であり、ウイルスの侵入および発芽過程を標的とした次世代抗インフルエンザ薬の開発に向けて強力な物質を創製できた。本物質の大量合成も試み、最終産物の前段階までこぎ着け、動物実験が出来る段階に来ている。本化合物は新規な抗インフルエンザ薬への応用が強く期待される。 2 Ru錯体のα-グルコース末端にシアロオリゴ糖鎖が1個及び2個転移したルテニウム複合体((YDS)-Ru conjugate)を創製した。本研究で、この物質がインフルエンザウイルスと結合すると特異的に蛍光が減少することを発見した。本物質はインフルエンザウイルス特異的に検出する新しい分子センサーとして大いに期待できる。 3 ヒトおよび動物由来の全てのインフルエンザA、B型ウイルス、全てのA亜型ウイルスに結合し、感染を阻害する全く新規なシアロスフィンゴ糖脂質(ガングリオシド)を発育鶏卵しょう尿膜中に発見し、部分構造を明らかにした。本物質は、インフルエンザウイルスの自然界における内因性のレセプターである可能性が極めて高く、この誘導体は、強力且つ画期的抗インフルエンザ薬として開発可能であることを見いだした。 4 いくつかの天然(分枝脂肪酸を含有するホスファチジルイノシトール他、2種の中性のグリセロ糖脂質)および人工化学合成した化合物(シアリルラクト系糖鎖を含む人工ムチン、シアリルラクトースを含む環状ペプチドなど)がインフルエンザウイルスの感染を阻止することを見出した。
