水溶性セレン試剤の開発とタンパク質化学への応用

研究概要	ジスルフィド結合(SS結合)をもつタンパク質のフォールディング試剤としては通常、水溶性イオウ試薬が用いられている。本研究ではイオウよりも酸化還元活性が高いセレンを用いることによって、従来のイオウ試剤とは異なる新しいタイプのタンパク質フォールディング試剤を開発し、これを用いて種々のタンパク質のフォールディング過程を詳細に解析することを計画した。同時に、有機化学的な視点(即ち、原子レベルの相互作用)からタンパク質立体構造の支配因子について考察するために、タンパク質構造データベース(PDB)の解析や非経験的分子軌道法による理論計算も行った。本年度はまず、これまでに合成した水溶性セレン試剤とタンパク質との反応速度論解析を行うために、(株)ユニソクの協力の下、速い反応過程(50ms〜1s)を追跡することができるクエンチフロー装置を設計・開発した。昨年度までの研究成果から、セレン試薬とタンパク質との反応は瞬時(1秒以内)に進行することがわかっている。装置は1月中旬に完成し、以降、タンパク質としてウシ膵臓リボヌクレアーゼAを用いて、フォールディング過程の詳細な解析を行った。50〜100msの間にタンパク質とセレン試剤との反応によってSS結合が生成してくる様子を、フォールディング中間体をイオン交換HPLCで分離することによって観測することに成功した。タンパク質フォールディングの初期過程の解析は現在注目されているトピックスである。本研究で開発したセレン試剤とクエンチフロー装置を用いることによって,フォールディングのごく初期に単寿命で存在する中間体の補足と観測を行える可能性があるものと考えている。PDBの解析からは、タンパク質立体構造の支配因子としてのS…O相互作用の存在を見出した。また、理論計算によって単一アミノ酸の構造特性が二次構造の形成に特に重要な役割を果たしていることを突き止めた。
