キノン光センサー複合体の構造・機能解析と光シグナリング経路の解明

研究概要	単細胞生物の光行動に関わるキノン光センサー複合体は、キノン色素類に属するブレファリスミンと200kDタンパク質が複合体を成した受容体であり、ロドプシンとは全く異なる新しいタイプの光センサーである。本研究は、キノン光センサーを介した未知の光シグナル変換/伝達経路を解明することである。まず、ブレファリスミン分子近傍(オルガネラの膜間部)の光依存的H^+濃度変化を解析・視覚化することによって、光受容オルガネラと細胞膜間のシグナル伝達がH^+によって仲介されていることを明らかにした(H^+濃度上昇によって膜の脱分極が起きる)。生化学的および分光学的解析の結果、キノン色素と200kDタンパク質が機能的複合体をなしていることがわかっている。しかし、なおも非特異的結合の可能性も完全には否定できない部分もあるため、本研究におい様々なアミノ酸やタンパク質との相互作用を分光学的手法を用いて解析した。この結果、キノン色素はいずれの場合も顕著な分光学的相互作用(クエンチング)を示すことはなかった。この結果から、200kDタンパク質は確かにキノン色素と機能的複合体をなしており、非特異的な結合ではありえないことが明らかになった。 200kDタンパク質の構造と光シグナル変換機能の分子機構を解明するために、タンパク質をコードする遺伝子のクローニングを10年前から試みている。しかしながら、200kDタンパク質を部分消化し純度の高いペプチド断片を得ることが究めて困難でありこの障害を突破することが当面の課題となっていた。本研究において技術的な問題点をほぼ解決し、ついに43kDペプチド断片の部分アミノ酸配列を決定することができた。この配列はGTQPNPSPASPVNであった。現在、さらなるペプチド断片を得て部分アミノ酸配列を決定し遺伝子クローニングに着手する予定である。
