線溶系関連遺伝子制御による虚血性神経細胞死の予防・防御に関する基礎的研究

研究概要	小動物を用いたヒトの病態に近い、神経細胞障害(細胞死:アポトーシス)モデルを確立し、これに対する線溶系因子の関与を検討した。平成13年度にはi)前脳虚血-再潅流にともなう海馬錐体細胞の遅発性神経細胞障害がアポトーシスであることを明らかにし、ii)このアポトーシス発現への線溶系因子の関与を明らかにする目的で、線溶系因子の一つである組織型plasminogen活性化因子欠損マウス(tPA-/-マウス)を用いて検討したが、wildタイプとの有意性は観察されず、関連性は否定的であった。iii)虚血-再潅流にともなう網膜神経障害発現においてはtPAの関与は否定的であったが、興奮性アミノ酸のNMDA刺激に伴うアポトーシス発現は、tPA-/-マウスにおいて有意に減弱し、線溶系因子の関与が示唆された。そこで平成14年度は、NMDA刺激に伴うマウス網膜へのアポトーシス発現への線溶系因子の関与をより詳細に検討した。 1)マウス眼内へのNMDA注入によるアポトーシス発現は、tPA-/-マウスではwildタイプに比べ、有意に減弱したが、これはtPAの眼内投与により解除された。2)今ひとつの線溶系因子である、ウロキナーゼタイプplasminogen活性化因子(uPA)欠損マウスならびに内因性のplasmin抑制因子(α2AP)欠損マウスにおけるNMDA刺激によるアポトーシス発現はwildタイプと差はなかった。3)plasminogen活性化因子に対する内因性の阻害因子(PAI-1)欠損マウスでのNMDA刺激に伴うアポトーシスはwildタイプに比べ有意に増大した。以上の結果から、内因性tPAは神経障害発現因子として関与していることが明らかとなり、また,その作用はtPAの線溶系活性とは異なる新たなメカニズムにより発現していることが強く示唆された。
