遺伝子ノックアウトによる膜結合型Prx4の精子形成における役割の解明

研究概要

ペルオキシレドキシン(Prx)はチオレドキシンならびにグルタチオンを電子供与体として過酸化物を還元無毒化する新規ペルオキシダーゼで、哺乳動物では現在6種類見つかっている。
1、その中の一つPrx6が肺や腎臓に強く発現し、酸化ストレスにより誘導されることから抗酸化活性に関係する可能性を示した。
2、Prx4は分泌シグナルをもつため血中に分泌されるが、精巣では切断されずに膜結合型として存在することが分った。抗Prx4抗体を用いた免疫組織染色にて検討したところ、精子形成細胞に強く発現していた。しかし、精子には本タンパクは存在せず、他の細胞質成分と共に遺残体中に検出された。免疫電顕で検討したところ、精子形成過程にある精子細胞の先体小胞ならびに遺残体内の小胞の膜系に強く認められた。手術により精巣を腹腔内にとどめる処理をした停留精巣モデルマウスでは、膜結合型Prx4のみが選択的に消失し、精子形成不全の病態に対応していた。遺伝子ノックアウトを行う目的でマウス・ゲノムライブラリーよりPrx4遺伝子をクローニングし、その構造をほぼ明らかにした。
3、Prxをはじめとするペルオキシダーゼはその活性にグルタチオンを必要とするが、酸化グルタチオンを還元再利用する酵素は主にセルトリ細胞に存在し、精子形成細胞にはほとんど検出されなかった。セルトリ細胞には酸化されたグルタチオンを還元して精子形成細胞に供給する働きがあることが分った。
