染色体変化の腫瘍内多様性からみたゲノム変化のtemporal analysis

研究概要	染色体DNAの1コピーの増減と2コピーの増減を識別するために、near-tetraploidのKATO-III細胞に正常細胞を種々の割合で混合したcontaminationmodelを作成してCGHを行い、混合比に応じたG/R比の変化がDOP-PCRによって修飾されるかどうかを検討した。その結果、CGHでは、DOP-PCRによる増幅の有無がG/R比のprofileや正常細胞の混合によるG/R比の変化に影響することはほとんどなかった。また、DNA-tetraploidの腫瘍では、理論値通り、正常細胞のcontaminationが20%未満の条件で1コピーのgainがCGHで捉えられた。染色体の1コピーlossを捉えることはCGHでは困難であったが、ゲノムDNAマイクロアレイでは1コピーlossの領域が部分的に捉えられた。この検討と平行して、Diffuse型胃癌23例にレーザ・マイクロダイセクションを用いて個々の腫瘍内の多数箇所からサンプリングを行い、抽出したDNAをDOP-PCRで増幅後、CGHに用いて染色体(部分)のコピー数変化を調べ、またゲノムマイクロアレイに用いてその変化の伽target geneを解析した。その結果から、個々の腫瘍内の検索したすべてのサンプルに見られた変化をstemline changesと定義し、進展の時間経過とともに、それらに更に変化(sideline changes)が付加していったプロセスを、多数のサンプルの結果から症例ごとに再構築した。また、検出された変化の中からearly eventsとlate eventsを抽出した。進行癌のearly eventsが早期癌の変化とどのくらい重なるのかを検討し、早期癌に見られて進行癌に少ない変化(4q+)、逆に進行癌に多く早期癌に少ない変化(7p+、15q+、3q-など)を明らかにした。
