研究概要

in situ RT-PCR法には増幅したcDNAの検出方法の違いから直接法と間接法があり、直接的in situ RT-PCR法は操作は簡便であるが非特異的標識や偽陽性所見が多く、一方で陽性所見に対する信頼性が高く非特異的標識が少ない間接的in situ RT-PCR法は、操作が煩雑となり実用性に乏しいとされている。われわれは直接的in situ RT-PCR法程度の簡便な操作で信頼性の高い結果の得られるin situ RT-PCR法の開発を試みている。この一年間でわれわれは、血管内皮増殖因子およびオステオポンチンメッセンジャーRNAに対するin situ RT-PCR法を施行し、その成果の一部は現在、英文学術雑誌に掲載予定である("Pancreas" in publication)。また、腫瘍特異的キメラ遺伝子の発現が特徴であるとされる軟部腫瘍のうち滑膜肉腫、末梢性神経外胚葉性腫瘍については、そのキメラ遺伝子メヅセンジャーRNAに対するin situ RT-PCR法を試み、その成果は既に複数の学会で発表済みである(第60回日本癌学会総会、第11回日本サイトメトリー学会総会)。in situ RT-PCR法の主な問題点である非特異的標識の回避についてはPCR反応条件の最適化、PCRプライマーの改良、nested PCRあるいはstep down PCR等の手法を併用することで解決を計りある程度の成果を得たが、さらなる検討が必要と考えている。検出感度については、TSA等の増感法の併用を試みる予定である。本年度新規に購入したサーマルサイクラーはin situ RT-PCR専用とすることで、研究の効率化、外来DNA混入の回避が達成された。
