腺管分離法を用いた大腸腫瘍のDNA multiploidの遺伝子解析

研究概要

我々は腺管分離法を用いて進行大腸癌のDNA ploidyと遺伝子異常との関係を検討してきた。それらの結果からDNA diploid(D)、aneuploid(A)、multiploid(M,D+A)は互いに異なるgenetic profileを有する独立したDNA typeであることを明らかにした(Sugai T, et al.Hum Pathol,2000;Sugai T, et al.Int J Cancer, 2000;Sugai T, et al.Oncogene,200;Sugai T, et al.Cytometry 2001;46:345-350.)。上記のDNA typeはDNA index(DI)からも異なっており、Aは1.3未満のlow DIで構成されているのに対して、Mは1.3以上のhigh DIで構成されていた。一方大腸腺腫のaneuploid stateのDIは1.3未満のlow DI腺腫と1.5以上のhigh DI腺腫に2大別されるが、早期癌の場合、腺腫内癌(group A cancer)と腺腫成分を含まないかもしくは極めて少量含む癌(group B cancer)に大別してDNA ploidy stateを検討した場合、前者の癌はlow DIで癌成分が構成されているのに対して、後者はlow及びhigh DI癌細胞で構成されていることが明らかになった(Sugai T et al.Pathol Int 2003;53:154-162)。上記の結果を基に、diploid、aneuploid、multiploidに基づいた大腸腫瘍の新しい分子腫瘍発生仮説を提案した(Sugai T, et al.J Pathol 2003;200:168-176)。その結果から、大腸腺腫のmultiploid化には18qが、早期大腸癌のmultiploid化にはp53遺伝子変異が関与していることが示唆された。
