βカテニン遺伝子変異ラット大腸粘膜細胞巣の腫瘍顕在化に関する分子病理学的研究

研究概要	研究は雄F344ラットにazoxymethaneの皮下注射により、大腸に比較的特異的に大腸腫瘍を形成できる大腸発癌モデルを使用して、大腸腫瘍を作成している途中である。経時的な屠殺により、それら腫瘍の前癌病変と考えられている従来からの大腸陰窩変異巣(aberrant cryptfoci ; ACF)と新規提唱されたβカテニン遺伝子変異巣(β-cantenin-accumulated crypt ; BCAC)を比較検討を行っている。がん予防物質候補であるシクロオキシゲナーゼー2(COX-2)の特異的抑制剤であるCelecoxib投与により、ACFは検索した大腸粘膜面積あたり陽性対照群で9.33土4.82に対してCelecoxib500ppm投与群で7.37±3.36(21%抑制)および1500ppm投与群で6.20士3.98(34%抑制)であった。一方BCACはそれぞれ4.77±1.55に対し、2.51±1.88(47%抑制)、1.45±1.25(70%抑制)であり、ACFの抑制率よりも有意に減少を認める結果を得、従来の前癌病変であるACFの抑制効果以上にBCACの抑制効果を示しており、このBCACがより前癌病変としてのパイオマーカーとなりうる可能性を示した。現在、前癌病変であるBCACを未処理の大腸粘膜から特定し得ないため、ホルマリン固定したパラフィン材料からのマイクロディセクション法での高率な抽出を試みている。また、未処理の大腸粘膜から特定方法の検討も始めている。
