βカテニン遺伝子変異ラット大腸粘膜細胞巣の腫瘍顕在化に関する分子病理学的解析

研究概要	研究は雄F344ラットにazoxymethaneの皮下注射により誘導されるβカテニン蓄積変異巣(beta-catenin-accumulated crypt ; BCAC)の前癌病変としての意義について、大腸がんのプロモーション作用を有する胆汁酸のひとつであるコール酸を利用して検討した。従来からの云われている前癌病変である変異陰窩巣(aberrant crypt foci ; ACF)との比較検討を行った。0.5%コール酸混餌投与により、ACFの頻度は大腸全体で65.1±23.1と対照群(194.9±23.2)に対し、有意に減少し、大きさも減少した。しかし、BCACは逆に対照群が2.21±0.99(単位面積あたり)に対し、2.60±1.05と増加傾向を認め、大きさも増大した。特に異常陰窩を6個以上集合する大きなBCACは有意に増加を示した。この結果はコール酸が大腸腫瘍発生の増加作用が示されていることから、ACFよりもBCACが大腸腫瘍形成に対するバイオマーカーとして意義か有ることを示唆している。現在、このBCACの病変から大腸腫瘍顕在化に関わる遺伝子の同定をmRNAレベルでの発現の違いを見ることで、検討しているが、mRNAレベルでの有意な差を示す遺伝子群は残念ながら、見つかっていない。その大きな原因の一つはACF及びBCACが顕微鏡サイズの大きさであるために、充分な量のmRNAの収集が困難であるためと考えている。今後の収集の効率化と方法論の改善をする予定である。
