皮膚ならびに粘膜組織におけるマウスランゲリン陽性細胞の動態と機能

研究概要	ランゲリンはヒト樹状細胞の一種である皮膚ランゲルハンス細胞に発現するC型レクチン分子である。本研究では、マウスを用いてランゲリンの詳細な解析を進めるため、昨年度マウスランゲリンcDNAのクローニングおよびFISHによる遺伝子マッピングを行い、またランゲリンmRNAの広範囲な発現および組み換えランゲリンのマンナン結合性等を明らかにした。これらをふまえ今年度は、1)ランゲリンmRNAは真皮と表皮の両者共にその発現が見られるが、ランゲリン抗体等によるマウス皮膚の免疫染色の結果、表皮ランゲルハンス細胞のみに蛋白の発現が確認されること、2)ヒトでは皮膚、扁桃、肺等に限られたランゲリン蛋白の発現が、マウスにおいては皮膚のみならず体表および腸間膜リンパ節、脾臓、胸腺等のリンパ器官に加え、肝臓等にも広く認められること、3)体表リンパ節、脾臓、肝臓については、これらの組織の中のCD8α^<high>CD11b^<low>樹状細胞サブセットのみに蛋白発現が限られること、4)ランゲリンのネックドメインに疎水性アミノ酸のheptad repeatsが存在し、蛋白架橋剤を用いた実験によって組み換えランゲリン分子が量体、四量体を経て多量体を形成すること、5)ランゲリン遺伝子導入細胞が糖蛋白質であるオボアルブミンを取り込むこと、6)同細胞が酵母細胞壁成分zymosanを捕捉すること等を明らかにした。以上の結果から、マウスにおいてはランゲリンはある種の樹状細胞特有の糖蛋白抗原・微生物取り込みレセプターとして働いている可能性が考えられる。
