多倍体化培養細胞株の樹立と多倍体化がもたらす形質変換の研究

研究概要

本研究の直接の目的は多倍体細胞を樹立し、多倍体化に伴う細胞形質の変化を求めるものである。3年間の研究で、4,6,8,10,12,16,18倍体Meth-A細胞及び3,4,6倍体V79細胞が樹立できた。この内、4,8,倍体Meth-A細胞と3倍体V79細胞が論文として報告されており、他は投稿中あるいは投稿準備中である。多倍化に伴う細胞形質の変化として、温度依存性、血清濃度依存性及び細胞膜糖鎖量が測定され、多倍体化に伴う細胞形質変化を観測することができた。本研究の目的は達成された。
本研究は細胞内DNA構造に関する既存の概念を修正しようとするパラダイムシフトを隠された目的の一つとしている。既存の概念では説明できない現象が本研究期間内で発見されている。2の冪乗倍体以外の細胞を樹立することができ、その倍数性は初期は暫時減少するが、その後は一定値で留まっている。このような現象は細胞内における染色体はほとんど独立していると考えている通説では説明できない。高橋学(故山口大学名誉教授)のDNA構造モデル(DNAフラクタル構造モデル)を多倍体細胞に拡張したモデル(ゲノムフラクタル構造モデル)を作成し、この現象を説明した(Cell prolif. 2002, 2003)。このモデルに基づく概念を広く知らしめるため、2冊の著書(1冊は高橋学の著書の翻訳本)を出版した。両書の要約は本研究の研究成果報告書(冊子体)に詳細を記載する予定である。
