c-kitミュータントを用いた肝再生機構の解析

研究概要	我々は、C-kitミュータント・Ws/Wsラットを用いて、肝細胞や肝幹細胞による肝再生の経路が傷害されたような状況下では造血幹細胞が肝再生を代償する可能性があることを示してきた。本年度はubiquitousにgreen fluorescent protein (GFP)が発現すると考えられているchicken β-actin promoter-GFPトランスジェニック(Tg)マウス[C57BL/6-TgN(act-EGFP)Osh1]の骨髄細胞をc-kitミュータント・W/W^vマウスに移植し、その肝臓におけるGFP陽性細胞の出現と分布を病理組織学的にしらべた。骨髄移植1ヶ月後の肝組織ではグリソン氏鞘周囲にGFP陽性細胞が出現し、四塩化炭素や抗Fas抗体を投与することにより肝障害を促すと類洞に沿ってGFP陽性細胞が観察されたが、肝細胞には明らかなGFP陽性像を検出することは難しかった。この実験系の結果は、移植した骨髄細胞の中には類洞内皮細胞へ分化できる細胞が存在することを示唆するものであるが、肝細胞への分化能については他の実験系で詳細な検討を加える必要性があると考えられる。今回ドナーに用いたC57BL/6-TgN(act-EGFP)Osb1マウスの肝細胞においてもGFP発現の検出が難しいことから、肝細胞に強く発現する遺伝子のpromoterで発現制御されるGFP-Tgマウスの作製の必要性について検討している。また、肝幹細胞や肝臓に生着した造血幹細胞に起源を発する多分化能細胞を単離するための新たなマーカーを探索している。
