c-kitミュータントを用いた肝再生機構の解明

研究概要

我々は、c-kitミュータント・Ws/Wsラットを用いて、肝幹細胞由来のoval細胞の発生には、c-kitレセプター・チロシンキナーゼ(KIT)を介したシグナルが重要な働きをしていることを明らかにしてきた。
胆管上皮細胞は、肝細胞と同様に肝芽細胞から発生するが、生後直ぐに成熟を完了する肝細胞とは異なり、しばらく未熟胆管上皮細胞として維持される。このような未熟胆管上皮細胞にも、KITが発現することが解ったので、Ws/Wsラットを用いて、未熟胆管上皮細胞の増殖におけるKITの生物学的作用について検討した。2週齢と6週齢のWs/Wsラットと正常(+/+)ラットの総胆管を結紮して、胆管上皮細胞の増殖を促した。結紮後4日目の肝組織標本を作製し、門脈領域(0.5x0.5mm^2)あたりのサイトケラチン(CK)-19陽性細胞数を算定するとともに、Ki-67抗原を指標としてその増殖能を検討した。2週齢では、+/+ラットの門脈領域に著しいCK19(+)-胆管上皮細胞の増加がみられたが、Ws/Wsラットでは、その増加は有意に抑制されていた。Ki-67抗原を指標とした細胞増殖能も、Ws/Wsラットでは+/+ラットに比べて有意に抑制されていた。一方、6週齢では、Ws/Wsラットと+/+ラットともに門脈領域にCK19(+)-胆管上皮細胞の増加がみられ、その増殖能も同程度であった。KITを介したシグナル伝達は、未熟胆管上皮細胞の増殖に重要な役割を果たしているのに対して、成熟した胆管上皮細胞の増殖には必須でないと考えられた。肝再生過程には、肝発生と共通したシグナル伝達機構が作用している可能性がある。
