ブラストシスチスのゲノム遺伝子の多様性を利用した人畜共通株の同定

研究概要	ブラストシスチスは1912年にヒトから見い出されて以来、種々の哺乳類・鳥類・爬虫類・両生類からも見い出されている。これら種々の動物由来のブラストシスチス株は、形態的に類似しているために、それらを区別・分類することは非常に困難である。一方、我々の1996年の研究成果から人畜共通性株がニワトリに存在することが判明したことから、形態的に区別困難なブラストシスチス分離株が人畜共通性なのか容易に判断できない状況にある。さらにヒトや動物由来のブラストシスチス株間には、遺伝子型の著しい多様性が知られており、これら多様な遺伝子型を調査すると、ヒトから分離・株化されたブラストシスチス株と同じ遺伝子型を示す株が、ヒト以外の動物由来のブラストシスチス株にも多数見い出されてきた。すなわち、このような株は人畜共通性のブラストシスチス株と考えられ、種々の動物がヒトへの感染源となっている可能性も考えられた。ブラストシスチスは、消化器症状を引き起こす可能性も指摘されており、感染源としての動物と人畜共通性のブラストシスチス株の同定、あるいは人畜共通性の株同定のための手法を確立することは重要なことである。本研究では、人畜共通性のブラストシスチス株同定のための簡便な遺伝子検索法を確立し、実際に各種動物から株分離を行った。昨年度に引き続き、人畜共通性株と判断されたブタ由来株やラット由来株から数種類の特異プライマーを構築した。さらに、家畜のブタやウシ、ならびに動物園で飼育されている霊長類や鳥類から人畜共通性のブラストシスチス株を多数検出し、特異プライマーの有用性を広く証明できた
