ブラストシスチスのゲノム遺伝子の多様性を利用した人畜共通株の同定

研究概要

近年ブラストシスチスはヒト以外の各種動物から高率に検出され始め、種名の問題と共に、ブラストシスチス分離株間の異同が議論されている。私達は、ヒトを含む種々の哺乳類だけでなく、鳥類、爬虫類、両生類からブラストシスチス株を分離し、それらの異同を分子生物学的手法によって解明し、さらに多種多様な動物由来株の近縁関係を明らかにし、ブラストシスチス感染はヒト・ヒト間だけでなく、動物・ヒト間で起っている可能性について検討してきた。本研究では、ゲノムの異同を区別するRAPD-PCR法を応用し、分離株間のゲノムの比較だけでなく、特有の遺伝子型のみを増幅するプライマーを構築した。このプライマーは非常に有効で、大阪市内の二ケ所の障害者施設内におけるブラストシスチス感染の伝播が施設利用者間で起きた可能性が大であることを証明した。さらに、プライマーによる増幅の有無により人畜共通性の遺伝子型株を簡便に同定できるシステムの開発と実際の応用について検討した。その結果、ヒト由来のブラストシスチス分離株間に認められる抗原性やゲノムの多様性が、動物由来株間にも同様に見い出され、かつ種々の哺乳類や鳥類由来のブラストシスチス分離株のほとんどはヒト由来のブラストシスチスと同じ遺伝子型であることが判明し、従って哺乳類と鳥類由来株の多くが人畜共通株であると想定された。この事は、動物に接触する機会が多い動物園関係者は、一般人よりもブラストシスチス感染率が高いという報告と一致することから、今後、感染動物が人への感染源として重要視されるべきであると考える。
