研究概要

研究代表者(吉田)は2001年7〜8月にかけて英国インペリアル大学において、海外共同研究者であるProf. Sinden, Prof. Crisantiらと共に本研究課題である『遺伝子導入によるマラリアを伝播しないハマダラカ作製』のための予備実験および研究打ち合わせを行った。Prof. Crisantiの研究グループと研究代表者は構築したプラスミドベクター(マラリア原虫の発育阻害をするタンパクをコード)を用い、すでに4系統のトランスジェニック蚊を樹立している。14年度はこれらトランスジェニック蚊のマラリア感受性を研究代表者がインペリアル大学において検討する。またProf. Crisantのグループは、新たなトランスジェニック蚊作製のための新技術(PiggyBac)を開発した(JBC 2002)。さらにハマダラカの強力なプロモーターとしてトリプシンプロモーターの解析を行い、trypsin regulatory elementを同定した(JBC 2001)。共同研究者であるProf. sindenのグループは、蚊のステージで重要な役割を演じているマラリア原虫のP25, P28, pbCHT1分子を同定し、knockout parasiteを作製してその機能を明らかにした(EMBO J 2001, Infect Immun 2001)。最近では、in vitro培養系でマラリア原虫のステージを再現することに成功した(Science 2002)。これらの研究成果は、トランスジェニック蚊研究の本課題の遂行に深く関わっている。
