遺伝子導入によるマラリアを伝播しないハマダラカの作製

研究概要

研究代表者(吉田)は海外共同研究者であるProf.Crisantiのグループと共同でハマダラカのカルボキシペプチターゼ遺伝子プロモーター領域をクローニングし、抗マラリア活性を有する一本鎖抗体遺伝子をこのプロモーター下に連結したプラスミドベクターを構築した。ハマダラカ卵にこのベクターDNAをインジェクションして4系統のトランスジェニック蚊を確立した。このトランスジェニック蚊は、吸血3時間後にはカルボキシペプチターゼ遣伝子プロモーターが活性化し、中腸内で一本鎖抗体遺伝子の発現を強力に誘導することを明らかにした。しかしながらこれらのトランスジェニックハマダラカにはマラリア伝播阻止能はなかった。現在中腸分泌型コンストラクトを持ったトランスジェニック蚊を作製中である。Prof.Crisantiのグループはハチ毒タンパクを発現するトランスジェニックハマダラカを作製し、これにマラリア伝播阻止能があることを見出した(JBC2002)。またトランスジェニックハマダラカによるマラリアコントロールの方向性を示す報告として、トランスジェニックハマダラカはWTに淘汰されないような工夫を施すことが必須であることを明確にした(Science2003)。海外共同研究者であるProf.Sindenのグループは熱帯熱マラリア原虫の各ステージにおけるタンパク発現プロファイルを明らかにした(Nature 2002)。またマウスマラリアゲノムプロジェクトにも参加し、その全塩基配列を明らかにした(Nature 2002)。
