新規動く遺伝因子(SCCmec)の起源とその転移機構

研究概要

市中獲得MRSA 3株(CA05、8/6-3P、81/108)のSCCmecの全塩基配列を決定し、これらの菌株のSCCmecは、typeII ccr遺伝子とclassB mec gene complex (IS1272-〓mecRI-mecA-IS431複合体)を持つ、新しいtypeのSCCmecであることを明らかにした。それぞれのJ-region(ccr gene complex及びmec gene complex以外の領域)の塩基配列も互いに異なるので、CA05のSCCmecをtypeIVa、8/6-3PのSCCmecをtypeIVb、81/108のSCCmecをtypeIVcSCCmecと命名した。また、メチリシン感受性CNSの標準株のなかで、S.hominis, S.arlettae, S.auricularisの標準株がccrA, ccrB遺伝子をもつことを見いだした。そのうちの一つS.hominis GIFU12263のccr遺伝子近傍の塩基配列を決定し、mec gene complexが存在しないが、ccrA1,ccrB1遺伝子をもつなどtypeI SCCmecと非常に良く似ていることを明らかにした。SCC_<12263>の存在は、SCCにmec gene complexが挿入されてSCCを形成するという考えを支持するものであった。S.hominisのccrA1,ccrB1及びtypeIV SCCmec上に存在するccrA2,ccrB2をクローニングし、このccr遺伝子が、部位特異的脱落と挿入をおこすことを確認した。TypeI及びTypeIV SCCmecはphage transductionで伝達されることを確認した。しかし、接合による伝達に関しては未だ研究途上である
