研究概要

Toll-like receptors(TLRs)は微生物構成成分を認識し、免疫系を活性化する一群の膜受容体である。ヒトでは10種類のTLRがこれまでに報告されている。各TLRは、細胞外のロイシンリッチリピート(LRR)ドメインで微生物成分を認識し、細胞内のTIRドメインを介してシグナルを伝達する。種々の微生物成分に対する免疫応答の相違は、TLRとそれに会合するアダプター分子の組み合わせに起因することが最近の研究より明らかとなってきたが、TLRによるリガンド認識機構に関しては未だ不明な点が多い。特に、TLR2はTLR1,TLR6,CD14などとヘテロダイマーを形成し、特殊な構造のLPSも含め多岐にわたる微生物成分を認識することが知られているが、リガンド認識及びシグナル伝達にco-receptor群がどのように関与するか明らかでない。本研究では、TLR2の細胞外領域の変異体を作製し、種々のTLR2リガンドを用いて認識に重要な領域の同定を行った。また、各変異体におけるTLR6とのヘテロダイマー形成能の有無を調べた。更に、我々が新たに同定したTLR3のアダプター分子であるTICAM-1(TIR-containing adapter molecule-1)のTLR2シグナリングへの関与を調べた。その結果、N末端に2分子の脂質を有するM.fermentans由来リポプロテインM161抗原及びそのリポペプチドMALP-2の認識には、TLR2のN末端領域が必須であること、M161抗原/MALP-2認識後のシグナルはTLR2単独の場合とTLR2/TLR6ヘテロダイマーの場合で異なることが判明した。また、TLR2及びTLR2/TLR6を介したシグナルは、MyD88,Mal/TIRAP依存的であるがTICAM-1非依存的であることを明らかにした。
