T細胞の運動制御における白血球特異的なパキシリン様蛋白質リューパキシンの意義

研究概要

【目的】leupaxinは、パキシリンと相同のドメイン構造をもち、T細胞をはじめとする造血系細胞に広く発現する。昨年度の研究を通じて、leupaxinはfocal adhesionに濃縮する特性をもち、integrin刺激依存的なパキシリンのチロシンリン酸化を抑制することを見いだした。本年度は細胞接着と運動制御におけるleupaxinの機能的な意義について以下の解析をおこなった。【方法】(1)leupaxinによる細胞接着制御:ヒト腎臓由来細胞293Tおよびヒト白血病細胞K562にleupaxinとEGFPの融合蛋白質を発現させ、fibronectinへの接着を解析した。(2)leupaxinの細胞進展に及ぼす影響:マウス線維芽細胞NIH3T3に野生型および種々の変異leupaxinを発現させ、細胞伸展に及ぽす影響を解析じた。(3)leupaxinのT細胞の運動制御における役割:マウスT細胞BW5147にleupaxinを発現させ、time-laps顕微鏡を用いて遊走するT細胞におけるleupaxinの分布と運動速度を解析した。【結果】(1)leupaxinはK562細胞のfibronectinへの接着を有意に抑制した。これに対して、293T細胞の接着には明らかな影響を与えなかった。(2)leupaxinはNIH3T3細胞のfibronectin上での細胞伸展を有意に抑制した。この抑制活性にはleupaxinのLD3モチーフおよぴLIM3ドメインが必要であった。(3)leupaxinはICAM-1上を遊走するBW5147細胞の後端部分に濃縮して存在した。一方、leupaxinの過剰発現のみではBW5147細胞のICAM-1上での運動速度に有意な変化は認められなかった。これらの結果から、leupaxinはintegrin基質への細胞接着や細胞伸展に対して抑制的に作用するが、その作用は細胞系譜に依存的であることが示唆された。また、遊走中のT細胞におけるleupaxinの分布は、これまでに報告されているパキシリンのそれとよく一致し、両者が協調的に作動してT細胞の運動を制御している可能性が示唆された。
