骨髄内及び骨髄外B細胞造血機構の解析

研究概要	本研究は骨髄内造血空間量により骨髄球系造血がB細胞系造血に優先して行われるという現象を利用して造血を制御する分子を明らかにすることを目的としている。 3種の原因の異なる破骨細胞欠損大理石病マウス[op/op、mi/mi、c-fos(-/-)]の、僅かに存在する骨髄腔では、B細胞系譜の造血のみが特異的に欠落していることを見いだした率統発生学的にも骨髄腔の発達に従って骨髄内造血は骨髄球、赤血球系譜がB細胞系譜に先行して行われる。B細胞造血に重要と考えられるSLF、Flt3/Flk-2 リガンド、IL-7、SDF-1等の遺伝子発現の差異を正常と大理石骨病マウスの骨髄で検討したが両者に変化は観察されなかった。そこでcDNAアレイ法を用いて、骨髄細胞由来の造血支持細胞株ST2の非刺激時と安定化β-カテニン遺伝子導入による発現の変化を検討したところ、多くの候補遺伝子が選出された。平成14年度ではこれらの分子の機能について検討する。マウス骨髄を除去すると著しい造骨が起こり、大理石病状態となりその後破骨細胞により骨髄腔が再形成される。本年度の計画どおり、正常マウスの骨髄除去術による骨髄再形成実験系も完成できた。次年度はこの実験系で骨髄除去によって全く造血が起こらない状態を作製し、再構築される造血環境の中で各血液細胞系譜の造血開始の順番を検討し、造血関連分子の発現の変化を検討する。我々の結果はD.Osmond, E.R.Stanleyという大御所にも注目され、彼らも同様の観察を最近報告した。また、サブトラクション法を用いてRANKLが増加しているとの結果も報告されてきた。
