Btkの新たな会合分子、BAM11の機能解析

研究概要	BtkはB細胞の抗原受容体、サイトカイン受容体やCD38等の機能分子を介するシグナル伝達に必須のチロシンキナーゼである。我々はBtkのPH領域に会合する分子BAM11(BAMと略す)'を単離し、BAMがBtkの活性化制御に重要な役割を果たすこと、BAMが転写促進活性を有することを報告した。今回、BAMの転写促進活性の制御機構について詳細な解析を試みた。 GAL4-BAM融合蛋白質発現遺伝子とGAL4結合DNA配列を上流につないだレポーター遺伝子を用いたルシフェラーゼアッセイの結果、BAMの転写促進活性の発現にはC末端領域が必須であることがわかった。また、Btkの共存によりBAMの転写促進活性はさらに上昇したが、PH領域欠損型、キナーゼ活性欠損型、およびxid型BtkおよびTec/BTKファミリーの一つであるltkでは活性の上昇はみられなかった。転写因子であるTFII-IはBtkと会合し、Btkが転写活性を亢進させることが報告されている。BAM、Btkと共にTFII-Iを発現させるとBAMの転写促進活性は更に増強されたが、Btkの非共存下ではTFII-IはBAMの転写促進活性に影響を与えなかった。BAMが酵母のTFG3/TAF30/ANC1と相同性を有すること、TFG3/TAF30/ANC1はクロマチンリモデリング因子であるSWI/SNF複合体の構成成分であり、INI1/SNF5と会合することから、免疫沈降法によりBAMとINI1/SNF5の会合を調べた。その結果、BAMとINI1/SNF5が会合することがわかり、SWI/SNF複合体の構成分子と相互作用することが示唆された。
