研究概要

ヒト精液に特異的なγ-glutamyl transpeptidase(γ-GTP)活性を示すユビキチン化CD10-CD13酵素複合体(SEGAP: seminal γ-GTP active protein)の生理的役割の解明を目的として、培養細胞内のmRNA発現の変化を観察した。さらに、inducible nitric oxide synthase(iNOS)の定量を目的としてiNOSのアミノ酸配列の一部に対するペプチド抗体を作製・検討した。
HL60およびHeLaを用い、SEGAPが細胞性免疫機構および子宮頚部細胞に及ぼす影響を観察した。培養細胞にSEGAPを添加後、経時的にRNAを精製し、RT-PCRを行った。サイトカインの中でIL-1β,IL-8,IL-12およびTNF-αはSEGAP刺激の有無にかかわらず発現しており、変化は観察されなかった。HL60ではiNOSがSEGAP添加15分後から発現したことから免疫抑制や抗菌・抗ウィルス作用が考えられた。MBL(mannose binding protein)はコントロールHeLaでは発現しないが、SEGAP刺激15分後にmRNAの誘導が観察されたことから、精子細胞表面に存在するマンノースサイトに細菌等が結合することによる感染からの防衛機能の可能性が考えられた。
iNOSのアミノ酸配列から2本の合成ペプチドを作製し、BSAに結合させた後に家兎に免疫した。得られた抗体はペプチドを結合させたSepharoseによりアフィニティー精製した。ヒトおよびマウスのリコンビナントiN0Sを抗原として抗体価を測定したところ、抗体1(10mg/ml)は主にヒトiN0Sに反応し、抗体2(4mg/ml)は主にマウスiNOSに反応し、いずれも1万倍希釈まで陽性であった。抗体の一部をビオチン標識し、サンドイッチELISAによるiNOS定量法の開発を進行中である。
