LPS識別におけるMD-2分子の機能解析

研究概要

グラム陰性菌の細胞壁構成成分であるLPS(lipopolysaccharide)は、あらゆる細胞に強い活性化を誘導し、臨床的にもしばしば致死的なショックを起こしエンドトキシンともよばれている。生体がどうやってLPSを認識するのかはこれまで不明であった。我々はMD-2分子を同定し、TLR(Toll-like receptor)4/MD-2複合体がLPS認識応答に必須であることをこれまで報告してきた。細胞株を用いた結果からMD-2がTLR4によるLPS認識に必須の分子である事を示してきたが、このMD2の重要性を正常免疫細胞、個体レベルで検討する為にMD-2KOマウスを作製した。MD-2が欠損したB細胞、マクロファージ、樹状細胞いずれもLPSに低応答性を示した。またエンドトキシンショックに対して抵抗性を示しSalmonella typhimurium感染に感受性が高かった。これらの結果からMD-2が個体レベルでもLPS応答に必須の分子である事が明らかとなった。MD-2のLPS応答における役割としてまず、TLR4の細胞内局在の制御があげられる。胎児由来線維芽細胞にTLR4を発現させ、その局在を調べたところMD-2の存在によってTLR4の局在が大きく変化する事を最近見出した。MD-2がないとTLR4はゴルジ体に停滞し、細胞表面への発現が認められなかった。
またMD-2はTLR4の局在ばかりでなくLPS認識そのものにも関与していると我々は考えている。LPS生合成前駆体であるlipidIVaは、マウスTLR4/MD-2にはアゴニスト、ヒトTLR4/MD-2にはアンタゴニストとして作用する。マウスTLR4/ヒトMD-2キメラ発現細胞には、lipidIVaはアンタゴニストとして作用した。このようにMD-2がマウスTLR4の特異性を制御することから、LPS認識・シグナル伝達にMD-2が直接関わることが明らかとなった。
